چکیده

Abstract B lymphocyte development is dependent on the interplay between chromatin landscape and lineage-specific transcription factors. It has been suggested that lineage commitment associated with major changes in nuclear environment, proposing a critical role for factors formation of epigenetic landscape. In this report, we have used chromosome conformation capture combination assay transposase-accessible sequencing analysis to enable highly efficient annotation both proximal distal transcriptional control elements genes activated specification mice. A large majority these were annotated at least one regulatory element an accessible configuration multipotent progenitors. Furthermore, binding sites key regulators specification, EBF1 PAX5, occurred already regions. did, however, cause dynamic change accessibility was increase promoter–enhancer interactions. Our data unravel extensive priming lineage-restricted provide insight into cell specification.

B lymphocyte lineage specification, commitment and epigenetic control of transcription by early B cell factor 1.

Early B cell factor 1 (EBF1) is a transcription factor that is critical for both B lymphopoiesis and B cell function. EBF1 is a requisite component of the B lymphocyte transcriptional network and is essential for B lineage specification. Recent studies revealed roles for EBF1 in B cell commitment. EBF1 binds its target genes via a DNA-binding domain including a unique 'zinc knuckle', which medi...

C/EBPα Is Required for Long-Term Self-Renewal and Lineage Priming of Hematopoietic Stem Cells and for the Maintenance of Epigenetic Configurations in Multipotent Progenitors

Transcription factors are key regulators of hematopoietic stem cells (HSCs) and act through their ability to bind DNA and impact on gene transcription. Their functions are interpreted in the complex landscape of chromatin, but current knowledge on how this is achieved is very limited. C/EBPα is an important transcriptional regulator of hematopoiesis, but its potential functions in HSCs have rem...

Notch and Nodal control forkhead factor expression in the specification of multipotent progenitors in sea urchin.

Indirect development, in which embryogenesis gives rise to a larval form, requires that some cells retain developmental potency until they contribute to the different tissues in the adult, including the germ line, in a later, post-embryonic phase. In sea urchins, the coelomic pouches are the major contributor to the adult, but how coelomic pouch cells (CPCs) are specified during embryogenesis i...

Pertussis toxin-sensitive G proteins regulate lymphoid lineage specification in multipotent hematopoietic progenitors.

Lymphoid and myeloid lineage segregation is a major developmental step during early hematopoiesis from hematopoietic stem cells. It is not clear, however, whether multipotent progenitors (MPPs) adopt a lymphoid or myeloid fate through stochastic mechanisms, or whether this process can be regulated by extracellular stimuli. In this study, we show that lymphoid lineage specification occurs in MPP...

Rapid lymphocyte reconstitution of unconditioned immunodeficient mice with non-self-renewing multipotent hematopoietic progenitors.

The replacement of abnormal hematopoietic stem cells (HSCs) with normal transplanted HSCs can correct a wide range of hematologic disorders. Here, we provide evidence that transplantation of more differentiated progenitor cells can be used to more rapidly correct lymphoid deficiencies in unconditioned immunocompromised mice. Transplantation of flk2+ multipotent progenitors led to robust B and T...
